Mycoplasma agalactiae PG2 et 5632 Whole genome shotgun
The genomes of the PG2 and 5632 strains of Mycoplasma
agalactiae each have a size of about 950 kb with a G+C% of about
30%. Genoscope will perform about 14,000 reads for each genome, in
order to obtain a depth of 10X, with an average read length of 650
bps. These reads are carried out using two types of libraries:
- Some prepared from products of mechanical shearing of the
genome (HydroSHear apparatus): one library from fragments of about
3 kb cloned in the pcdna2.1 vector (5,000 clones sequenced on both
ends) and another from fragments of about 10 kb cloned in the pcns
vector (1,000 clones sequenced on both ends).
- Another library whose inserts were obtained by partial
digestion of each genome by Sau3A. The fragments, of about 20 kb,
were cloned in the pBBc miniBAC vector; 1,000 clones from this
library will be sequenced on both extremities. The clone links
produced by these sequences will be useful in the assembly
phase. The finishing phases will produce a quality sequence which
conforms to the Bermuda rules.
