The objective of the project is to sequence an approximately 1-megabase genomic region of two genotypes of Phaseolus vulgaris (the common bean). These sequences will be compared with each other and to orthologous regions of several species of legumes in which the phylogenetic distance is variable.
For this project Genoscope is undertaking the sequencing of 20 BACs (10 BACS per genotype) situated in the region of the resistance gene Co-2 in two genotypes of Phaseolus vulgaris. Each BAC will undergo sub-cloning (6kb inserts cloned in pcdna2.1) followed by shotgun sequencing at 10X coverage
