Genoscope is undertaking the sequencing of the complete genome of the DM4 strain of Methylobacterium extorquens*, estimated at 7 Mb (based on the model strain AM1).
The genome will be sequenced at a depth of 12X, using three libraries:

  • a 3 kb insert library cloned in the multicopy plasmid vector pcdna2.1,
  • a 10 kb insert library in the pCNS plasmid vector (small copy number)
  • and a miniBAC library ( 30kb inserts) in the pBBC vector (derived from pBeloBAC11). Shotgun sequencing should produce about 120000 reads. A finishing step will increase the quality of the sequences obtained.

*First named Methylobacterium dichloromethanicum, the sequenced strain belongs to the Methylobacterium extorquens species.
